肿瘤抑制基因【肿瘤转移相关基因1与抑制基因KISS-1在胰腺癌中的表达及意义】

  [摘要] 目的 研究肿瘤转移相关基因1(metastasis associated 1,MTA1)与抑制基因KISS-1在胰腺癌中的表达,探讨其与病理指标的关系。 方法 应用免疫组化Envision二步法在36例胰腺癌组织、10例癌旁胰腺组织中检测MTA1与KISS-1蛋白的表达情况。 结果 MTA1在胰腺癌组织中的阳性表达率为77.8%(28/36),显著高于癌旁胰腺组织中的表达[20.0%(2/10)](P < 0.01);KISS-1在胰腺癌组织中的阳性表达率为38.9%(14/36),明显低于癌旁胰腺组织的90.0%(9/10)(P < 0.01)。二者的表达均与患者年龄、性别及肿瘤部位无关(P > 0.05),而与癌组织的分化程度、淋巴结转移及临床分期有关(P < 0.05)。胰腺癌组织中MTA1与KISS-1的表达呈负相关(r = -0.533,P < 0.01)。 结论 MTA1和KISS-1与胰腺癌的发生、侵袭以及转移有关,其有望成为胰腺癌诊治的重要生物学指标。

  [Abstract] Objective To study the expression of tumor metastasis associated 1 (MTA1) and suppressor gene KISS-1 in the pancreatic cancer and explores its relationship with the pathological indicators. Methods The immunohistochemical Envision two-step method was used to detect the MTA1 and KISS-1 protein expression in 36 cases of pancreatic cancer tissue and 10 cases of normal paracancer tissue. Results The positive expression rate of MTA1 in pancreatic cancer was 77.8% (28/36), which was significantly higher than the 20.0% (2/10) in the normal paracancer tissue (P 0.05), but related to the differentiation degree of cancer tissue, lymph node metastasis and clinical stage (P 30%为3分;显色强度:不显色或显色不清为0分,浅黄色为1分,棕黄色为2分,深棕色为3分。二项乘积0~1为阴性(-),2~3为弱阳性(+),4~5为中度阳性(++),> 5为强阳性(+++)。
